Recent years have seen an increasing interest in metabolic engineering of C. glutamicum for production of organic acids. Here we report a new engineered strain capable of lysine production which has 10-fold high lysine production capacity than the wild type strain. In this book we provided further insight into AK regulation and function and also provided valuable information for engineering L-lysine producing C. glutamicum strains. The data provided in this book show the importance of the deregulation of aspartate kinase and site directed mutagenesis for overproduction of L-lysine in C. glutamicum. The new engineered strain in this study promises to create new opportunities in bioprocess performance and over produce L-lysine for therapeutic and biomedical applications. The recombinant strain described here may provide a basis for future metabolic engineering to obtain a further increase in lysine production by C. glutamicum.
