The current study was undertaken to investigate the role of macrophages as a cellular immune function against immunization with whole sonicated Listeria monocytogenes antigens (WSLMAgs) and the effect of probiotics. The results revealed the cellular immune function against pre- and post-immunization in spleen organ via lymphocytic hyperplasia in white pulp and coalescence of lymphoid follicles and marker F4/80+ show the immune-positive cells in aggregation adjacent to lymphoid follicle or focal aggregation of macrophages between follicles as well as marker F4/80+ show the immune-positive cells (brown color) in sinusoids of the liver. The outcome proved a statistically noteworthy difference (P <0.05) between the WSLMAgs have able to increase the amount of Interleukin-12 (IL-12) in comparison to the probiotics group and control group due to able to induce the immune response of innate and acquired cells as well as a humoral response after challenge with virulent Listeria monocytogenes. The Effectiveness of ultra-sonicator LM pre and post-immunization then challenge with virulent LM in the induction of cellular immune response might serve as an immunization platform for applicants
