The study aimed to isolation and identification of Potato virus Y Egyptian isolate (PVY-EG) using biological serological and molecular characterization as well as study the sensitivity of some potato cultivars to this isolate infection for select one cultivar or more containing PVY resistant gene. On the other hand the available information about the effect of PVY on potato plants physiology is not studied yet. Therefore this study was carried out on potato plants infected with PVY-EG to clarify the role of the PVY-EG (pepper-Fayoum) in potential risk or actual damage to the potato growth it has been necessary to study the pathogenicity effects of PVY-EG isolate thoroughly physiological impact. It also touched on this study to the biochemical and molecular properties of these cultivars since were gel electrophoresis of total proteins using the SDS-PAGE technique as well as isozyme analysis of POX and PPO in addition to research in cultivars that resistant to PVY in different eleven cultivars then nucleotides sequencing of the Ryadg gene in both these cultivars were performed.
